    Penelitian ini bertujuan untuk mengetahui Seberapa besar pengaruh ladder drill terhadap kecepatan memanjat atlet panjat tebing kota Balikpapan. Penelitian ini merupakan penelitian kuantitatif dengan metode eksperimen. Teknik pengambilan data dalam penelitian ini menggunakan data pre-test dan data post-test. Populasi dalam penelitian ini seluruh atlet panjat teing kota Balikpapan yang aktif berlatih dengan menggunakan sampling purposive, dengan jumlah 15 orang. Analisis data menggunakan analisis deskriptif dengan persentase, dan batuan SPSS Versi 26. Berdasarkan hasil penelitian secara keseluruhan dari 15 orang atlet yang mana pada Berdasarkan perolehan data pretest, rata-rata waktu pemanjatan adalah 13,80 detik dengan waktu tercepat 8,12 detik dang paling lambat 22,55 detik, sementara untuk rata-rata waktu dari data posttest yaitu 9,99 detik dengan waktu tercepat 7,04 detik dan waktu paling lambat 15,58 detik.hal ini ditunjukan dengan nilai t hitung= 6,186 >  t tabel = 2,145 dan nilai signifikansi p sebesar 0.000 < 0.05 dengan persentase peningkatan 27,60%.     Effect of Regional Variation on the Total Flavonoid Level of Ethanol Extract of Mangosteen (Garcinia Mangostana) Peels

    Background : Currently, traditional medicines have been widely used by the public. One of them is mangosteen peel. Extract of mangosteen peel contains alkaloids, glycosides, steroids, flavonoids, polyphenols, and tannins. In order to ensure the quality of the extract so that its chemical content can be guaranteed, it is necessary to standardize the quality of the extract that consists of specific parameters. Objective : This study aims to determine the specific parameters using Thin-Layer Chromatography (TLC) and total flavonoid level of the ethanol extract of mangosteen peel from Kalimantan, Java, and Sumatra. Methods : This research was an explorative study. Extraction technique used was maceration with 70% ethanol solvent. The tested specific parameters, namely the identity of the extract with TLC and total flavonoid level, were determined afterward using visible spectrophotometry with the AlCl3 reagent. Results: The result of the qualitative test with TLC showed that the extract of mangosteen peel contains flavonoids, terpenoids, and anthraquinones. Total flavonoid level of ethanol extract of mangonsteen peel from Kalimantan, Java, and Sumatra were (0.301 ± 0.009); (0.398 ± 0.015); (0,747 ± 0,010) mg QE/g extract, respectively. Conclusion : Extract of mangosteen peel contains flavonoids, terpenoids, and anthraquinones.     The CD38/NAD/SIRTUIN1/EZH2 Axis Mitigates Cytotoxic CD8 T Cell Function and Identifies Patients with SLE Prone to Infections

    Summary: Patients with systemic lupus erythematosus (SLE) suffer frequent infections that account for significant morbidity and mortality. T cell cytotoxic responses are decreased in patients with SLE, yet the responsible molecular events are largely unknown. We find an expanded CD8CD38high T cell subset in a subgroup of patients with increased rates of infections. CD8CD38high T cells from healthy subjects and patients with SLE display decreased cytotoxic capacity, degranulation, and expression of granzymes A and B and perforin. The key cytotoxicity-related transcription factors T-bet, RUNX3, and EOMES are decreased in CD8CD38high T cells. CD38 leads to increased acetylated EZH2 through inhibition of the deacetylase Sirtuin1. Acetylated EZH2 represses RUNX3 expression, whereas inhibition of EZH2 restores CD8 T cell cytotoxic responses. We propose that high levels of CD38 lead to decreased CD8 T cell-mediated cytotoxicity and increased propensity to infections in patients with SLE, a process that can be reversed pharmacologically. : Katsuyama et al. find that an expanded CD8CD38high T cell population in SLE patients is linked to infections. CD8CD38high T cells display decreased cytotoxic capacity by suppressing the expression of related molecules through an NAD+/Sirtuin1/EZH2 pathway. EZH2 inhibitors increase cytotoxicity offering a means to mitigate infection rates in SLE.     Virtual Team Leader Communication:Employee Perception and Organizational Reality

    Based on a study of leader communication effectiveness conducted in a large human resource outsourcing firm, this article reports how virtual team members’ perceptions of their leaders’ effective use of communication tools and techniques affect team performance outcomes. The study also investigates the role that trust plays in moderating the relationship between virtual team members’ perceptions of their leaders’ effective use of communication and team performance. Analysis of 458 responses from 68 teams found a positive relationship between virtual team members’ perceptions of leaders’ effective use of communications and team members’ perception of their team’s performance. The study also found that trust strengthens the relationship between perceived leader communication effectiveness and team performance results. Last, the study also revealed serious organizational alignment issues between what team members perceived to be effective leader communication, their perception of team performance outcomes, and the organizations performance measured by a balanced scorecard

    Variation at the vernalisation genes Vrn-H1 and Vrn-H2 determines growth and yield stability in barley (Hordeum vulgare) grown under dryland conditions in Syria

    The objective of the present study was to identify QTL influencing agronomic performance in rain-fed Mediterranean environments in a recombinant inbred line (RIL) population, ARKE derived from the Syrian barley landrace, Arta and the Australian feed cultivar, Keel. The population was field tested for agronomic performance at two locations in Syria for 4 years with two sowing dates, in autumn and winter. Genotypic variability in yield of the RIL population was mainly affected by year-to-year variation presumably caused by inter-annual differences in rainfall distribution. The spring growth habit and early flowering inherited from the Australian cultivar Keel increased plant height and biomass and improved yield stability in Syrian environments. QTL for yield and biomass coincided with the map location of flowering time genes, in particular the vernalisation genes Vrn-H1 and Vrn-H2. In marginal environments with terminal drought, the Vrn-H1 allele inherited from Keel improved final biomass and yield. Under changing climate conditions, such as shorter winters, reduced rainfall, and early summer drought, spring barley might thus outperform the traditional vernalisation-sensitive Syrian landraces. We present the ARKE population as a valuable genetic resource to further elucidate the genetics of drought adaptation of barley in the field

    Resolution, Relief, And Resignation:A Qualitative Study Of Responses To Misfit At Work

    Research has portrayed person–environment (PE) fit as a pleasant condition resulting from people being attracted to and selected into compatible work environments; yet, our study reveals that creating and maintaining a sense of fit frequently involves an effortful, dynamic set of strategies. We used a two-phase, qualitative design to allow employees to report how they become aware of and experience misfit, and what they do in response. To address these questions, we conducted interviews with 81 individuals sampled from diverse industries and occupations. Through their descriptions, we identified three broad responses to the experience of misfit: resolution, relief, and resignation. Within these approaches, we identified distinct strategies for responding to misfit. We present a model of how participants used these strategies, often in combination, and develop propositions regarding their effectiveness at reducing strain associated with misfit. These results expand PE fit theory by providing new insight into how individuals experience and react to misfit—portraying them as active, motivated creators of their own fit experience at work

    Mutual trust between leader and subordinate and employee outcomes

    Stable and enduring cooperative relationships among people are primarily based on mutual trust. However, little evidence exists about the effects of mutual trust between supervisor and subordinate on work outcomes. To understand better the dynamics of trust in supervisor–subordinate relationships, we examined how mutual trust between supervisor and subordinate is associated with work outcomes. Based on a sample of 247 subordinate–supervisor pairs, multilevel analyses revealed a positive effect of perceived mutual trust on task performance and interpersonal facilitation after controlling for trust in leader and felt trust. In addition, task performance and interpersonal facilitation increased as trust in leader and felt trust or trust in subordinate both increased

    Using Light to Improve Commercial Value

    The plasticity of plant morphology has evolved to maximize reproductive fitness in response to prevailing environmental conditions. Leaf architecture elaborates to maximize light harvesting, while the transition to flowering can either be accelerated or delayed to improve an individual's fitness. One of the most important environmental signals is light, with plants using light for both photosynthesis and as an environmental signal. Plants perceive different wavelengths of light using distinct photoreceptors. Recent advances in LED technology now enable light quality to be manipulated at a commercial scale, and as such opportunities now exist to take advantage of plants' developmental plasticity to enhance crop yield and quality through precise manipulation of a crops' lighting regime. This review will discuss how plants perceive and respond to light, and consider how these specific signaling pathways can be manipulated to improve crop yield and quality

    Isolation in Globalizing Academic Fields: A Collaborative Autoethnography of Early Career Researchers

    This study examines academic isolation – an involuntary perceived separation from the academic field to which one aspires to belong, associated with a perceived lack of agency in terms of one’s engagement with the field – as a key challenge for researchers in increasingly globalized academic careers. While prior research describes early career researchers’ isolation in their institutions, we theorize early career researchers’ isolation in their academic fields and reveal how they attempt to mitigate isolation to improve their career prospects. Using a collaborative autoethnographic approach, we generate and analyze a dataset focused on the experiences of ten early career researchers in a globalizing business academic field known as Consumer Culture Theory. We identify bricolage practices, polycentric governance practices, and integration mechanisms that work to enhance early career researchers’ perceptions of agency and consequently mitigate their academic isolation. Our findings extend discussions on isolation and its role in new academic careers. Early career researchers, in particular, can benefit from a deeper understanding of practices that can enable them to mitigate isolation and reclaim agency as they engage with global academic fields
